Multi-Omics Profiling Alters Treatment Recommendations in 76% of Ovarian Cancer Patients: TPC Study in Nature Communications

Ovarian Cancer Tumor Profiling Resource Published in Nature Communications

The Tumor Profiler Consortium celebrates the publication of a comprehensive multi-omics resource for high-grade serous ovarian cancer (HGSOC) in Nature Communications. The study demonstrates that integrated multimodal tumor profiling can inform personalised maintenance treatment decisions and reveals how chemotherapy reshapes tumour cell diversity.

The prospective, multi-centre Swiss TuPro gynaecological project profiled 87 samples from 62 patients using up to eleven technologies spanning DNA, RNA, protein and functional assays — analysing blood, solid tumour tissue and malignant ascites within a four-week turnaround time suitable for clinical decision-making.

Key Findings

• Multi-omics data altered hypothetical treatment recommendations for 76% of patients compared with routine care and standard genomic analysis alone
• Multi-omics-guided maintenance therapy was associated with prolonged overall survival in newly diagnosed patients
• Whole-genome amplification, present in 43% of samples, emerged as a predictor of earlier disease recurrence and altered ex vivo drug response
• Chemotherapy exposure coincided with a marked increase in patient-specific cancer cell heterogeneity and genomic signature alterations

The authors conclude that molecularly guided treatments should be tested as adjuvant therapies prior to chemotherapy in future trials. These findings have already informed the prospectively randomised OV Precision Trial (Swiss-GO Trial Group / Tumor Profiler Center, NCT06466382), which is currently enrolling newly diagnosed ovarian cancer patients.
